Some guy on GameFAQs picked up the Prima Stradegy guide for DMC4 today and is giving out alot of details I am going to post alot of them below. Read at your own risk.
Story Spoilers (Boss List and Final Boss, Also Neros TRUE Idenity)
Final Boss : The last Boss is *Drum Roll* Sanctus, with the Sparda sword no less, than he gets possessed by the Giant Statue, and you go at it one final time, the last time is more of a story fight kinda like the last Mundus fight in DMC.
Neros Idenity : Nero's Identity isn't reveled but rather alluded to. As I mentioned above, almost all of the order enemy's names have Angelo in them and most of the enemies are constructed from pieces of Nelo Angelo's soul. Take that as you will.

Secret missions and Missions : There are 12 secret missions. Blue orb fragments for completing them. Some as Dante some as Nero.
You play as Dante from mission 12 to mission 18. 19 and 20 are Nero's endgame.
Unlockables
Bloody Palace is a battle arena mode similar to the same bonus mdoe from DMC3 SE. The player battles through 101 stages, the player can choose to play it as Nero Super Nero, Dante or Super Dante in Normal or automatic mode.
Every 10 stages there is a boss, at stages 10, 30, 50, 70, 90 the enemies are grouped difficulty enough to be considered bosses. Stage 99 is also done in this manner. in stages 20, 40, 60, 80 and 100 you fight story bosses like Berial. Stage 101 is Dante in quote "His most powerful and dangerous form"

*** and *** mode(Same as Heaven or *** except Nero/Dante only takes one hit to die but enemies can take alot more than one hit)

The unlockables are on the short side (Unless the guide is hiding something)
Beating Human gives you The history of DMC video, Game Clear Art: The Two Heroes and a set of 29 character images.
Beating Devil Hunter gives you all the same as above plus Game Clear Art: The cast, Son Of Sparda mode and the bloody palace.
Beating Son of Sparda mode gives you Bonus Clear Art: The Demons, 12 Publicity pictures, DMD mode, HOH mode.
Beating DMD gives you Game Clear Bonus Art: Demon Invasion, *** and *** mode, Super Nero and Super Dante.
HoH mode gives you Bonus Art: The ladies of DMC,
*** and *** mode gives you Bonus art: Light from the Demon Blade.
Also, While Son Of Sparda mode is essentially hard mode there is one change. In order to progress to the next level you MUST get at least an A rank.
